Introduction A controllable lysis system was engineered in Synechocystis sp. PCC 6803 to enhance lipid extraction for biofuel production. Bacteriophage-derived lysis genes were integrated into the genome and activated by a nickel-inducible signal transduction system. Three strategies were employed: using phage lysis cassettes, expressing endolysin genes while restricting holin genes, and combining lysis genes from different phages. This system significantly induced autolysis in Synechocystis cells with NiSO4 addition, offering an alternative to mechanical or chemical cell breakage and facilitating biofuel recovery from cyanobacteria.
