Introduction In this study, new piano-stool ruthenium(II) complexes were synthesised to assess steric hindrance effects of bulky phosphane ligands on halide exchange reactions. Using η6-arene rings such as methyl benzoate and p-cymene, four complexes were created, but they exhibited poor cytotoxicity initially. However, aged DMSO solutions of one complex demonstrated improved activity over time. This led to the discovery that certain complexes transform in DMSO to cyclometalated species, enhancing cytotoxicity. Cyclometalated complexes were synthesised and tested, showing greater activity compared to non-cyclometalated counterparts.
