Oxymatrine Powder Description

Oxymatrine Powder (C15H24N2O2) is a white or off‐white crystalline solid. It has a bitter taste and no distinct odour. It exists as a stable, non‐hygroscopic material under standard conditions. It gradually darkens upon prolonged light exposure. The compound shows moderate thermal stability. Decomposition occurs near 210°C without a distinct melting point. Its molecular structure contains a quinolizidine nucleus, an N‐oxide group and a bridged oxygen atom. These features contribute to both polarity and conformational rigidity. The powder shows high solubility in water (approximately 1:8 w/v) and in lower alcohols such as methanol and ethanol. It dissolves poorly in non‐polar solvents such as chloroform or ethyl acetate. Crystalline analysis reveals an orthorhombic morphology. It exhibits characteristic infrared absorption bands at 1640 cm⁻¹ (carbonyl), 1100 cm⁻¹ (N→O stretch) and 2850–2980 cm⁻¹ (alkyl groups). UV–Vis spectroscopy indicates maximum absorption at 220 nm. The powder remains stable in acidic conditions. It degrades gradually in alkaline environments (pH > 9), particularly at elevated temperatures. Its crystalline lattice permits polymorphism. The alpha form is the most common. Particle size distribution typically ranges from 10–100 micrometres. The bulk density is near 0.6 g/cm³. Optical rotation measurements yield [α]D20 = +30° (c = 1, H₂O). The N‐oxide group confers zwitterionic properties. It allows salt formation, and the bicyclic system contributes to stereospecific bioactivity. Common Preparation Methods

Oxymatrine powder is industrially extracted from the dried roots of Sophora flavescens using a multi‐stage process. The crushed roots are subjected to acidic aqueous extraction using dilute sulphuric or hydrochloric acid at pH 2–4 and temperatures of 80–90°C for several hours to solubilise alkaloids. The acidic extract is then basified to a pH of 9–10 with sodium hydroxide or ammonia, precipitating crude alkaloids that are collected by filtration. The crude mixture undergoes solvent partitioning with chloroform or dichloromethane to remove impurities. Matrine is subsequently oxidised to oxymatrine using hydrogen peroxide under controlled conditions. The converted oxymatrine is purified through recrystallisation from ethanol–water systems or preparative chromatography. The resulting crystals are washed with a cold solvent and vacuum‐dried at 40–50°C, then milled into a uniform white powder. Final quality control includes HPLC purity verification (>98%), a melting point check (decomposition near 210°C) and residual solvent testing to comply with pharmacopeial standards.
