Tris Base Description

Tris Base crystallizes as hygroscopic orthorhombic needles or free-flowing powder with a molecular weight of 121.14 g/mol and decomposition at 219°C. The tertiary amine structure enables effective buffering across pH 7.0–9.2 (temperature coefficient: -0.031 pH/°C), with zwitterionic behavior in aqueous solutions. It demonstrates high solubility in water (55 g/100mL at 25°C), formaldehyde, and DMSO, but limited solubility in ethanol (<0.5 g/100mL). Thermal gravimetric analysis confirms stability below 150°C, with decomposition releasing trimethylamine and aldehydes above 219°C. Pharmaceutical grades maintain stringent controls: heavy metals (Pb ≤0.5 ppm), chloride ≤0.005%, sulfate ≤0.002%, and endotoxin ≤0.05 EU/mg for cell culture applications. UV absorbance at 280 nm is ≤0.03 AU in 1 M solutions.

CO₂ absorption forms carbonic acid, lowering pH. Always store solutions under inert gas; prepare fresh monthly. Powder remains stable >3 years if sealed.

Q2. Can Tris replace HEPES in cell culture?

Only for short-term applications (<24 hr). Tris lacks HEPES' constant pKa across 15–37°C and may inhibit some cell lines at >25 mM.

Q3. How to avoid metal contamination in DNA workflows?

Use nuclease-free grades (DNase/RNase undetectable). Chelate residual metals with 0.1 mM EDTA if required.

1.       Common Preparation Methods

Tris Base is synthesized via condensation of nitromethane with formaldehyde under alkaline conditions, followed by catalytic hydrogenation (Raney nickel, 80–100°C). Crude crystals undergo triple recrystallization from deionized water/ethanol (3:1 v/v) to remove residual amines and metals. The purified solution is treated with activated carbon, filtered through 0.22 μm membranes, and vacuum-concentrated. Crystallization occurs at 4°C with seeding to control polymorphism. Crystals are centrifuged, washed with cold acetone, and fluid-bed dried at 45°C under nitrogen to achieve ≤0.1% moisture. Final milling produces powder with 90% particles 50–200 μm.
