Wogonin (CAS: 632-85-9) is an O-methylated flavon, a flavonoid-like chemical compound found in Scutellaria baicalensis. The glycosides of Wogonin are known as Wogonosides. Oroxindin, for example, is a Wogonin-glucuronide isolated from Oroxylum indicum. It is one of the active ingredients in Sho-Saiko-To, a Japanese herbal supplement.

Wogonin (5,7-Dihydroxy-8-methoxyflavon) is an O-methylated flavon from the flavonoid category, primarily derived from Scutellaria radix. Wogonin exhibits anti-inflammatory effects by suppressing the expression of iNOS and COX-2. Additionally, Wogonin has potential benefits in inflammatory colorectal carcinogenesis, primarily through the suppression of NF-κB and the activation of Nrf2 signalling pathways in HCT116 cells and THP-1 cells, which have been used for the treatment of pneumonia. Its anti-inflammatory properties are linked to the inhibition of NO, cytokines, chemokines, and growth factors in dsRNA-induced macrophages via the calcium-STAT signalling pathway. It has been found that Wogonin can mitigate the impairment of cardiomyocytes induced by hyperglycaemia through anti-inflammatory and antioxidant mechanisms. Furthermore, Wogonin significantly inhibited PPAR-γ mediated phosphorylation and activation of NF-κB-P65. Overall, Wogonin demonstrates anti-inflammatory effects by suppressing inflammatory mediators, including IL-6, COX-2, PGE2, iNOS, and NO in IL-1β-stimulated osteoarthritis chondrocytes[1].

Studies have shown the inhibitory effects of Wogonin on various cancer cells. The mechanisms include modulation of the p53 signalling pathway, slowing G1 phase progression, antitumor angiogenesis through inhibition of VEGF, and apoptosis via the mitochondrial pathway. These findings suggest that Wogonin is a novel anti-cancer agent, enhancing therapeutic effects on chemotherapy-resistant tumours, and could be clinically beneficial for patients with ovarian cancer. Polier et al. demonstrated for the first time that Wogonin and structurally related natural flavones, such as Apigenin, Chrysin, and Luteolin, inhibit cyclin-dependent kinase 9 (CDK9) and block the phosphorylation of the carboxy-terminal domain of RNA polymerase II. This effect leads to reduced RNA synthesis and consequently a rapid downregulation of the short-lived anti-apoptotic protein Myeloid Cell Leukaemia 1 (Mcl-1), resulting in the induction of apoptosis in cancer cells.[2]
